ABSTRACT

Docetaxel that causes microtubule stabilization during mitotic spindle organizing in cell and forces the cell to go apopitosis is a widely used drug in breast, prostate and bladder cancers. Cytotoxic effect of docetaxel which has been used in the treatment of many other cancers on multiple myeloma cell line (RPMI- ATTC No: CCL-155) was investigated with different drug concentrations. Metastatic breast cancer [MCF-7 (human breast cancer) ATCC No: HTB-22] (human breast cancer) cell line was used as the control group. Cytotoxic effects occurring with increasing docetaxel concentrations in cell cultures of RPMI-8226 and MCF 7 cell lines were evaluated by XTT assay. Cytotoxic effect caused by docetaxel was lower in MCF-7 (human breast cancer) cell lines of the control group. However cytotoxic effect with increasing docetaxel concentrations in RPMI-8226 (human multiplee myeloma) cell lines was demonstrated. Data demonstrate that docetaxel with a concentration of 50 μmol reaches a lethal dose on RPMI-8226 cell lines. Low cytotoxic effect of docetaxel in the control group is only explained by the metastatic character of breast cancer cell line. Efficient cytotoxic effect of docetaxel in multiple myeloma cell line suggests that this drug may be used in the treatment of this disease. However further animal experiments are needed for the more efficient use of docetaxel in the treatment of multiple myeloma.
