Microbiologic Analysis of the Spring Water Sources at the Military Unit in Ankara With Polymerase Chain Reaction Method

In the present study, well water sources of military troops in Ankara Garnison were analyzed with a new Multiplex Polymerase Chain Reaction (PCR) method which is developed by using current PCR methods used in microbologic analyses of water.

28 spring water samples were collected from four different military units in Ankara Garnison. We used Multiplex PCR method for thermotoleran (total) coliforms, Escherichia coli, Salmonella and Shigella analysis in water samples.

We found E.coli in samples taken from well waters in the rate of 50%. We did not find Salmonella and Shigella in water samples. Standart bacteriological methods for the examination of water generally take about 48 hours and generally only one type of bacteria is investigated, but the whole procedure in this study for four bacteria took about 4 hours. For these reasons, the new Multiplex PCR method presented in this study, will be very useful especially in the situations like outbreak, flood and earthquake, which the investigation of the water sources is urgent.
